چکیده
Institute for Molecular Genetics, Baylor College of Medicine, Houston, Texas 77030 Ever since it was shown that PCR could simultaneously amplify multiple loci in the human dystrophin gene, (1) multiplex PCR has been firmly established as a general technique. A short list of multiplex PCR applications now includes pathogen identification, gender screening, linkage analysis, forensic studies, template quantitation, and genetic disease diagnosis. Multiplex PCR can be a two-amplicon system or it can amplify 13 or more separate regions of DNA. It may be the end point of analysis, or preliminary to further analyses such as sequencing or hybridization. The steps for developing a multiplex PCR and the benefits of having multiple fragments amplified simultaneously, however, are similar in each system. This review focuses on multiplex systems in which each primer pair targets a single locus, unlike RAPD (2) and alumorph {3) PCR reactions, which amplify multiple loci with a single primer or primer set.

Department of Pathology, State University of New York at Stony Brook, Stony Brook, New York 11794 Since the discovery of the structure of DNA in 1953, a great deal of effort has been devoted to detecting specific target sequences. The first commonly used technique, filter hybridization, offered an excellent sensitivity of one copy per 100 cells. Restriction endonuclease digestion in conjunction...
متن کاملDevelopment of Multiplex Reverse Transcription-Polymerase Chain Reaction for Simultaneous Detection of Influenza A, B and Adenoviruses
Background and objective:Millions of people in developing countries lose their lives due to acute respiratory infections, such as Influenza A & B and Adeno viruses. Given the importance of rapid identification of the virus, in this study the researchers attempted to design a method that enables detection of influenza A, B, and adenoviruses, quickly and simultaneously. ...
متن کاملDevelopment of a Multiplex RT-PCR Assay for Detection of the Causal Agents of Citrus Tristeza and Cachexia Diseases with Coamplification of Plant mRNA as an Internal Control
Background and Aims: Plant certification programs need reliable, fast, cheap and sensitive methods for detection of systemic pathogens with special interest in virus and viroid detection. Reverse transcriptase-polymerase chain reaction (RT-PCR) has been documented as an alternative assay for certification of plant propagating materials. The main object of the present study was the optimization ...
متن کاملInternal and External Controls for Reagent Validation
PCR applications that require a high confi dence in the result should be designed to control for the occurrence of false negatives. False negatives can occur from inhibition of one or more of the reaction components by a range of factors. While an external, or batch control is often used, the ideal control is one that is included in the reaction cocktail in a multiplex format. Early approaches ...
متن کاملDevelopment of a multiplex polymerase chain reaction assay for differentiation of field strain isolates and vaccine strains S19 and RB51 of Brucella in Iran
Bovine brucellosis is a zoonotic disease distributed worldwide and characterized by abortion and reducedfertility in cows. Since brucellosis eradication programme in Iran uses vaccination, test, slaughter andquarantine as control measures, it is essential to distinguish vaccine strains from strains that cause infectionsamong vaccinated cattle herds. We developed and evaluated a multiplex polyme...
